A: Effect of increasing concentrations of N-acetylcysteine (NAC) on the disappearance of PDX-1 measured by Western blot analysis using nuclear extracts of HIT-T15 cells cultured in RPMI media containing either 0.8 or 11.1 mmol/l glucose for passage 74 (p74; control) and passages 123–126 (p123-126). N-acetylcysteine prevented the disappearance of PDX-1 protein that was associated with prolonged culturing with the higher glucose concentration. B: Effect of increasing concentrations of N-acetylcysteine on DNA binding of PDX-1 measured by electromobility shift assay of nuclear extracts in the same passages of HIT-T15 cells as described in A. N-acetylcysteine prevented the disappearance of PDX-1 binding that was associated with prolonged culturing with the higher glucose concentration. The methods are the same as in Tanaka et al. (12).
